Title Determination of Lactobacilli and Bifidobacteria in Commercial Products containing Lactic Acid Bacteria

  • Abstract Lactic acid bacteria in commercial products were determined by means of plate-counting method. Two commercial products consisting of lactobacilli and bifidobacteria, as indicated on the labels, were assayed in this work. After a serial dilution by a factor of 100, ten and a hundred microliters of the diluted suspensions were spread on MRS agar plates. These were then incubated at 30?C for 48 hours. The plates incubated in anaerobic jar have more colonies than that cultured under aerobic condition. The viable count of product (I) were 1.9×10^7 cfu/g which is 1/10^4 fewer than that indicated on its label. The viable count of product (II) were 7.2×10^8 cfu/g which approximated one tenth as it indicated. The colonies were picked up and cultured in MRS medium and then the lactic acid and acetic acid in the culture broth were determined by HPLC. Product (I) contained lactobacilli and is free of bifidobacteria based on acetic acid to lactic acid ratio. In contrary, product (II) has solely bifidobacteria.
