The aim of the thesis is to culture these five lines of adventitious roots under different cultural factors: different root length and wounding, inocula and subculture periods in order to obtain the adventitious root line with high growth ratio and flavonoids production. Among different culture test, T05 line ( 3-4 cm root length and 0.06 g/10 ml inoculation) expressed the highest growth ratio (27.81±1.4) and total flavonoids content was up to 86.48±5.97 eq. iridin mg/g DW and the total of isoflavonoids content was 45.83±1.22 mg/g DW after four weeks culture. In bioreactor culture, T05 line has a highest growth ratio (13.49±0.56) and total flavonoids content (48.31±1.48 eq. iridin mg/g DW), when culture in modified MS medium with 2% sucrose and 1 g/100 ml inoculums. The content of total fiavonoids and isofiavonoids of T05 line was increased when cultured in MS medium added with methyl jasmonate. The growth ratio and total flavonoids content of T05 line and wild type adventitious roots were compared. The growth ratio of T05 line is up to ten folds higher than the wild type in 2L bioreactor culture. The total flavonoids content of T05 line is two folds higher than rhizome growing in the field.
