Title Studies on micropropagation and secondary metabolites content of several strawberry (Fragaria × ananassa) cultivars

  • Abstract   For germination rate test, fourteen sources of strawberry seeds were cultured in sterile water or on Murashiage and Skoog (1962) medium without plant growth regulator after surface sterilization. The results showed that seeds soaked in sterile water have higher germination rate than those cultured on medium. However, the rate of germination was from 17.54 to 98.63 %, which depends on different sources of seeds. The runner or shoots from Taoyuan No.4, Perfume, Toyonoka and Large Toyonoka were used as explants, which cultured on MS basal salts containing different concentrations of antioxidants, Polyvinylpyrrolidone (PVP), and added individually or in different combinations with growth regulators for shoot multiplication. The best response was the Toyonoka’s runner shoots, multiple shoots were formed when cultured in 1/2MS basal medium supplemented with 5 mg/l PVP ,1.0 mg/l Thidiazuron (TDZ) and 0.5 mg/l Indole-3-butyric acid (IBA) after four months. Callus and adventitious shoots were regenerated from the edge of Toyonoka leaf explants cultured on 1/2MS basal medium supplemented with 5 mg/l PVP, 1 mg/l TDZ and 0.1 mg/l 2,4-Dichlorophenoxyacetic acid (2,4-D) after 2 weeks.
      Methanol extracts from fourteen samples of strawberry were analyzed. The shoots and leaves from field-grown Toyonoka’s plant expressed highest content of total flavonoids (20.256 eq. quercetin μg/mg DW) and total phenols (130.298 eq. gallic acid μg/mg DW),and the amount of catechin, epicatechin, procyanidin B1 and procyanidin B2 were 20.739μg/mg DW, 2.967 μg/mg DW, 4.953 μg/mg DW and 8.563 μg/mg DW, respectively.
